摘要:
      目的:探讨补肾中药对破骨细胞(osteoclast,OC)骨吸收功能的影响。方法:取新生(出生24h内)SD乳鼠四肢骨髓分离培养OC,实验组加入不同(高、中、低)剂量的补肾中药血清,对照组采用无中药血清,采用酶动力学法测定培养上清中抗酒石酸酸性磷酸酶(tartrate resistant acid phosphatase,TRACP)的活性;利用甲苯胺蓝对骨吸收陷窝染色并在图像分析仪下测定骨吸收陷窝的面积和数目。结果:补肾中药高、中剂量组均降低TRACP活性,减少骨片上骨吸收陷窝的面积及数目,与空白对照组比较差异有显著性意义(P<0.01),高、中剂量组间比较差异有显著性意义(P<0.05),以中剂量组作用为明显。结论:补肾中药可降低破骨细胞培养上清中的TR ACP活性,减少骨吸收陷窝面积及数目,达到抑制破骨细胞的骨吸收功能目的。

Abstract:
      Objective:To investigate the kindney-tonifying TCM efficacy on bone resorption by cultured osteoclasts of rats in vitro.Method:The culture medium containing the serum from rats fed with the drugs at different dosage(high, moderate, low dosage) were used to treat the osteoclasts from the bone marrow of newborn SD rats,and the tartrate resistant acid phosphatase(TRACP) and the bone resorption pits were observed. TRACP activities in the medium were measured by enzyme kinetics. The bone resorption pits were stained by toludineblue. The area and number of resorption pits were determined with Leica Quantimet 500 system. Result:Kindney-tonifying traditional Chinese drugs at high and moderate dosage significantly inhibited the TRACP activities, decreased the area and the number of bone resorption pits, as compared with the control(P<0.01). The effect at the moderate dosages was much better than high dosage, which showed significant difference between them(P<0.05).Conclusion:Kindney-tonifying traditional Chinese drugs can inhibited the function of resorption of osteoclasts.
