摘要:
      目的探讨早期"强制性使用"对局灶性脑梗死大鼠大脑皮层微管相关蛋白-2(MAP-2)表达的影响.方法70只Wistar大鼠随机分为假手术(D)组(10只)和手术组(60只),后者制作成右侧大脑中动脉阻塞(MCAO)模型,24h后再分为患侧前肢固定(A)组、未固定(B)组和健侧前肢固定(C)组,每组20只.21天后解除固定,于MCAO后21天和28天时,用免疫组化的方法检测梗死灶周围大脑皮质MAP-2的表达.结果与D组相比,手术组大鼠梗死灶周围皮质MAP-2阳性神经元数量明显减少,但光密度值增高(P<0.05或P<0.01);手术组大鼠梗死灶周围皮质MAP-2阳性细胞数及光密度值,B组高于A组,A组高于C组(P<0.05).结论脑缺血损伤可刺激MAP-2的表达,肢体固定或过度使用对梗死灶周围皮质部分神经元代偿性高表达MAP-2有不利影响,其中患侧前肢过度使用的影响尤其明显.

Abstract:
      Objective:To study the effect of early forced use on expression of MAP-2 in the peri-ischemia cortex after local cerebral infarction.Method:70 rats were randomly divided into: sham-operated group (group D n=10) and operated group (n=60). 60 right middle cerebral artery coagulated Wistar rat were divided into 3 groups at random: Contra (to the lesion)antevion eotremity cast(group A), No cast(group B) and Ipsi cast (group C). The cast was used at 24 hours after MCAO and removed at day 21 postsurgery. The expression of MAP-2 in the cortex of the three groups were evaluated at 21d and 28d after the infarction.Result:In operated group the number of MAP-2 labeled cells decreased significantly, but the expression of MAP-2 increased, in comparison with group D (P<0.05,P<0.01). The number of MAP-2 labeled cells and the expression of MAP-2 in group B were higher than those in group A and group C(P<0.05,P<0.01);those in group A were higher than those in group C(P<0.05).Conclusion:The cerebral infarction could induce the expression of MAP-2. Immobilization of UE, especially immobilization of unaffected-UE (forced use affected-UE), inhibited expression of MAP-2 in the peri-ischemic cortex.
