摘要:
      目的:探讨低氧与运动联合应用对大鼠肺血管重构及结缔组织生长因子(CTGF)表达的影响,揭示低氧/缺氧适应机制。方法:30只雄性SD大鼠随机分为常氧对照组(C组)、低氧组(H组)和低氧与运动联合组(H+E组)。H组与(H+E)组置于模拟海拔3000m,8h/d, 6d/周, 5周;(H+E)组辅以低负荷跑台训练,5°,15m/min,1h/d。测量肺动脉平均压(mPAP)、血管(WT)%、(WA)%,RT-PCR检测CTGF mRNA表达变化情况。结果:H组与(H+E)组大鼠mPAP、WT%、WA%及CTGF mRNA均较C组明显升高(P<0.05);(H+E)组大鼠上述指标则显著低于H组(P<0.05)。结论:低氧与运动联合应用降低CTGF表达可能是减轻肺血管重构的重要原因,CTGF可能与低氧/缺氧适应密切相关。

Abstract:
      Objective: To investigate the effects of hypoxia-exercises on pulmonary vascular structural remodeling and expressions of connective tissue growth factor(CTGF). Method: Thirty male SD rats were divided into control group, hypoxia group and hypoxia-exercises group. The (mPAP), (WT)% and (WA)% were measured. The expressions of CTGF mRNA in pulmonary arteries were evaluated by reverse transcriptase-polymerase chain reaction (RT-PCR) technique. Result: The mPAP, WT%, WA%, and CTGF mRNA level in hypoxia group and hypoxia-exercises group were significantly higher than those in control group(P<0.05), while those in hypoxia-exercises group were significomtly lower than those in hypoxia group(P<0.05). Conclusion: After hypoxia-exercises CTGF mRNA level reduced, which may result in ameliorating pulmonary vascular structural remodeling. CTGF may play an important protective role in the mechanism of hypoxic altitude acclimation of pulmonary tissue.
