摘要
目的:探讨“白蛋白-突变型肌肉生长抑制素前肽”融合蛋白在抗肌萎缩中的作用。
方法:首先将肌肉生长抑制素前肽基因第76位天冬氨酸对应的碱基突变为丙氨酸对应的碱基,获得了“突变型肌肉生长抑制素前肽”基因。再将该基因与大鼠白蛋白基因片段连接,克隆至酵母表达载体。利用酵母表达系统,重组表达“白蛋白-突变型肌肉生长抑制素前肽”融合蛋白。纯化的重组蛋白用于尾悬吊大鼠的治疗实验。
结果:大鼠经过21天的尾悬吊,后肢骨骼肌发生了明显的萎缩。注射“白蛋白-突变型肌肉生长抑制素前肽”融合蛋白的尾悬吊大鼠,其后肢骨骼肌未发生萎缩。
结论:重组表达的“白蛋白-突变型肌肉生长抑制素前肽” |

Abstract
Objective:To investigate the effect of ‘myostatin propeptide-albumin’ fusion protein on preventing muscle atrophy.
Method:Aspartate-76 to alanine-76 mutant myostatin propeptide gene was prepared and then linked to rat albumin gene segment. This fusion gene was cloned into yeast expression vector to generate recombinant fusion protein . Expressed fusion protein was purified, then supplied to tail-suspended rats.
Result:After 21d of tail suspension, hind limb muscle atrophy could be observed in tail suspension rats, whereas the atrophy could be prevented in the tail suspended rats by injection of ‘myostatin propeptide-albumin segment’.
Conclusion:Recombinant fusion protein ‘myostatin propeptide-albumin’can inhibit muscle disuse atrophy caused by muscle unloading. It may be a potential therapeutic application for muscle disuse atrophy. |
