目的:通过电针足三里和阿是穴,研究急性骨骼肌钝器伤后,电针治疗对转化生长因子-β1(TGF-β1)和波形蛋白表达的影响,探讨电针抑制纤维化和促进肌纤维再生的机制。
方法:将60只新西兰兔随机分为模型组(A组)、阿是穴组(B组)、足三里组(C组)、足三里+阿是穴组(D组)4组,每组15只。电针组在造模后24h治疗,隔日1次。于造模后第7、14、28天取材,运用两步法免疫组化检测TGF-β1和波形蛋白。
结果:与A组相比,各电针组于骨骼肌损伤后(第7天和第28天)TGF-β1表达显著下降(P<0.05),D组表达低于B组和C组,B组低于C组(P<0.05),第14天时,B组与A组无明显差异(P>0.05)。波形蛋白在A组中少量表达,且随着时间延长呈下降趋势,而电针组中在第14天呈上升趋势,B组和C组波形蛋白表达低于D组(P<0.05),但仍高于A组(P<0.05),B组波形蛋白表达低于C组,但差异没有显著性(P>0.05)。
结论:电针有抑制骨骼肌纤维化、促进骨骼肌再生的作用,可能与抑制TGF-β1和提高波形蛋白的表达水平有关;电针刺激阿是穴或足三里均有利于受损骨骼肌的修复且两穴具有协同作用。 |

Abstract
Objective:To explore the effects of electroacupuncture on expressions of transforming growth factor-β1(TGF-β1) and vimentin and the mechanism of inhibiting fibrosis and promoting muscle fibers regeneration after acute blunt contusion in rabbits.
Method: Sixty rabbits were randomly divided into model group (group A,n=15),Ashixue group (group B,n=15),Zusanli(ST36) group(group C,n=15),Ashixue+Zusanli group(group D,n=15).Electroacupuncture groups were treated within 24h after contusion once every other day. Immunohistochemical method was used to detect TGF-β1 and vimentin on the 7th d, 14th d and 28th d after modelling.
Result:As extension of time after injury, the expressions of TGF-β1 in electroacupuncture groups declined significantly compared with group A. The expressions of TGF-β1 in group D was the lowest and had differences with other electroacupuncture groups(P<0.05). The level of vimentin in group A showed a downward trend over time. But the trend was contrary on the day 14 and showed upward trend among three treatment groups.The results showed that group D had a potence of increasing the expression of vimentin and the expression was lower in group B and group C(P<0.05), but which was still significantly higher than group A(P<0.05). Expressions of vimentin in group B and group C had no statistical significance.
Conclusion:The mechanism of electroacupuncture on inhibiting the skeletal muscle fibrosis and promoting skeletal muscle regeneration may be related to inhibiting the expression of TGF-β1 and raising the expression of vimentin;electroacupuncture at Ashixue or Zusanli acupoints are all beneficial for repairing the damaged skeletal muscle, and the effects of these two acupoints are synergic. |
