目的:探讨“形神共养”的丰富生存环境对大鼠缺血再灌注脑损伤后神经功能及突触超微结构的影响。
方法:将60只雄性SD大鼠随机分为缺血“神养”组(IE1)、缺血“形养”组(IE2)、缺血“形神共养”组(IE3)、缺血标准环境组(IS)、假手术标准环境组(SS)、正常对照组(NC),每组10只。制作大鼠缺血再灌注模型。IE1、IE2、IE3组均给予丰富生存环境训练;其余组居于标准环境。于第3天、10天、17天、24天、31天进行改良神经功能缺损评分,实验结束后透射电镜观察缺血侧皮质和海马的突触超微结构的形态变化。
结果:于训练第10天、17天、24天、31天,IE1、IE2和IE3组的行为学评分有不同程度的改善,且与IS组比较有显著性差异(P<0.05)。患侧皮质和海马IE1、IE2、IE3组突触后致密物厚度、突触间隙宽度与IS组相比有显著性差异(P<0.05),且IE2、IE3两脑区突触界面曲率与IS组相比有显著性差异(P<0.05)。
结论:“形神共养”组大鼠的丰富生存环境训练对脑缺血再灌注的神经功能改善有促进作用。 |

Abstract
Objective: To study the effects of "preservation from both physique and spirit" of enriched environment on the recovery of neurological function and synaptic structures morphology after focal ischemia/reperfusion in rats.
Method: Sixty male SD rats were randomly divided into an ischemia + preserving the spirit enriched environment group (IE1), an ischemia+preserving the physique enriched environment group (IE2), an ischemia + preservation from both physique and spirit enriched environment group (IE3), an ischemia+standard environment group (IS), a sham operation+standard environment group (SS), a normal control group (NC), 10 rats in each group. Rats in the ischemia/reperfusion groups had their middle cerebral artery sutured for two hours before reperfusion. IE1,IE2,IE3 groups were put into three kinds of enriched environment cages. The others were put into the standard cages without any training. Modified neurological severity scores(mNSS) was used to evaluate the neurological function of rats at the 3rd, 10th, 17th, 24th, 31st days after operation. Synaptic structures in cortex and hippocampus area at ischemia side were assessed morphologically and quantitatively.
Result: The scores of mNSS were significantly better than IS group (P<0.05) after 1 week training. The post-synaptic density (PSD) increased significantly and the synapse cleft width were narrower in the three enriched environment groups as compared with the IS group (P<0.05).The synaptic curvatures improved obviously in the IE2 and IE3 groups in contrast to the IS group (P<0.05).
Conclusion: The “preservation from both physique and spirit” enriched environment group can promote the functional recovery in rats after cerebral ischemia injury. |
