目的:研究丰富环境对阿尔茨海默病模型SAMP8小鼠海马CA1区炎症反应的影响。
方法:将20只5月龄雄性SPF级SAMP8小鼠随机均分为P8+标准环境(SE)组和P8+丰富环境(EE)组两组,分别在SE和EE下喂养60d,将10只5月龄雄性SPF级SAMR1小鼠作为对照组,于SE下饲养60d,称为R1+SE组。通过免疫组织化学染色检测各组小鼠海马CA1区胶质纤维酸性蛋白(GFAP)和促炎细胞因子,包括白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)的表达。
结果:P8+EE组小鼠海马CA1区GFAP、IL-1β、IL-6和TNF-α的平均吸光度明显少于P8+SE组小鼠(P<0.01),但仍显著高于R1+SE组(P<0.01)。
结论:丰富环境干预可抑制阿尔茨海默病模型SAMP8小鼠海马CA1区星形胶质细胞(AST)的活化、增生,降低促炎细胞因子释放,减轻炎症反应,这可能是丰富环境改善阿尔茨海默病脑功能的作用机制之一。 |

Abstract
Objective: To investigate effects of enriched environment on inflammatory reaction in hippocampal CA1 region in senescence accelerated mouse prone/8 with Alzheimer’s disease.
Method: A total of 20 5-month-old male SPF SAMP8 were randomly divided into P8+SE group and P8+EE group with 10 in each group. Mice in the two groups were respectively subjected to standard environment and enriched environment for 60 days. A total of 10 5-month-old male SPF SAMR1 mice were selected as control group (referred to as R1+SE group) and were subjected to the standard environment for 60 days. The expression of glial fibrillary acidic protein (GFAP) and proinflammatory cytokines, such as inter leukin-1 beta (IL-1β), inter leukin-6 (IL-6), tumor necrosis factor alpha (TNF-α) in hippocampal CA1 of mice in each group were determined by immunohistochemical staining.
Result: The average absorbance of GFAP, IL-1β, IL-6 and TNF-α in P8+EE group were obviously less than those in P8+SE group (P<0.01), but they were still significantly higher than those in R1+SE group (P<0.01).
Conclusion: Enriched environment could inhibit the activation and proliferation of astrocytes (AST), reduce the release of proinflammatory cytokines and inflammation in hippocampal CA1 region in senescence accelerated mouse prone/8 with Alzheimer’s disease, which may be one of the mechanism that enriched environment improve brain function of Alzheimer’s disease. |
