      摘要 目的:探讨功能性电刺激(FES)治疗改善脑梗死大鼠的神经功能是否与缺血半暗带GSK-3β蛋白磷酸化水平的变化相关。 方法:大鼠随机分为假手术组、对照组及FES治疗组,每组均分为治疗0天(治疗前)、3天、7天、14天四个亚组。建立大鼠永久性大脑中动脉栓塞模型(pMCAO)并进行皮下电极埋置。术后3天开始行FES治疗,每天1次,每次30min。在各时间点对大鼠进行改良神经功能损害评分(mNSS)评定神经功能,随后取材,用Western Blot检测半暗带GSK-3β蛋白磷酸化水平的变化。 结果:治疗第7、14天FES治疗组大鼠mNSS评分明显低于对照组(P<0.01),FES治疗组治疗第7天和第14天的大鼠神经功能较第0天和第3天有明显改善(P<0.05)。治疗第7天,GSK3β磷酸化水平FES治疗组高于对照组和假手术组(P<0.05),而FES治疗组治疗第7天的GSK-3β磷酸化水平明显高于第3天和第14天(P<0.05)。 结论:FES能通过增高GSK-3β磷酸化水平改善脑梗死大鼠的神经功能。

      Abstract Objective: To discuss whether the alteration of neurological impairment of rats with cerebral infarction after the treatment of functional electrical stimulation(FES) was relevant to the phosphorylation of glycogen synthase kinase-3β(GSK-3β) in their ischemic penumbra fields. Method: Totally 72 rats were randomly divided into three groups: sham-operation, control and NMES treatment. They were subdivided into 4 subgroups, 0-day(before treatment), 3-day, 7-day, 14-day treatments, and 6 rats in each. Cerebral infarction was achieved by the permanent middle cerebral artery occlusion(pMCAO). NMES treatment with hypodermic electrodes was provided once per day and 30 min per time. Modified neurological severity score (mNSS) and phosphorylation of GSK-3 in brain were assessed at each observation point. Result: For the mNSS points and comparing with the control group, the improvement of neurological function was observed among rats treated for 7 and 14 days (P<0.05), but not in 3 days treatment. As the phosphorylation of GSK-3β in the rats penumbra field, it is higher in control group and NMES group than those in sham operation group, and the highest is the 7days treatment group. Conclusion: NMES can ameliorate the neurological impairment after cerebral infarction, and its mechanism involves elevating the phosphorylation of GSK-3β.
