      摘要 目的:对比分析低强度全身振动与阿仑膦酸钠防治去势大鼠骨质疏松效果。 方法:104只SD大鼠随机分为对照组(sham组)、卵巢摘除术+注射生理盐水组(OVX+VEH组)、卵巢摘除术+全身振动组(OVX+WBV组)和卵巢摘除术+阿仑膦酸钠组(OVX+ALE)。OVX+VEH组是单纯卵巢切除后给予生理盐水(1mg/kg,每周灌胃1次);OVX+WBV组是切除卵巢后给予全身振动(振动强度为0.3g,频率为45—55Hz,每次振动20min,每天1次,每周5次,休息间隔不大于2d,共12周);OVX+ALE组是切除卵巢后给予阿仑膦酸钠(1mg/kg,每周灌胃1次);定期收集标本,测定血清中骨钙素(osteocalcin, OC)和Ⅰ型胶原C端交联肽(CTX)水平,micro-CT检查分析和三点弯曲生物力学检测。 结果:血清中OC和CTX在卵巢切除后均增加显著(P<0.001),WBV和ALE治疗组对OC影响不大,但ALE能够显著抑制血清CTX的增加。在实验12周时,与WBV治疗组比,ALE治疗组更能够显著提高胫骨近端松质骨的骨微结构参数[BV/TV(+25.3%, P=0.004), TV apparent (+60.3%, P<0.001), BV material (+28.4%, P=0.044), Tb. N (+15.6%, P=0.001), and Tb. Th (+16.9%, P=0.037)]。在实验12周时,与OVX+WBV治疗组对比时,OVX+ALE治疗组能够提高胫骨的最大载荷、骨的刚度参数和骨的吸收能量,但差异无显著性意义。 结论:相对于低强度全身振动,阿仑膦酸钠能够更好的防治去势大鼠骨质疏松和改善骨的微结构。

Abstract:
      Abstract Objective:To investigate the different effect between low-magnitude whole-body vibration(WBV) and alendronate(ALE) on bone properties in ovariectomized rats. Method: A total of 104 SD rats were randomly divided into four groups (sham, OVX+VEH, OVX+WBV, OVX+ALE). The level of WBV applied was 0.3g at 45—55Hz for 20 min/d, 5d/week and for 12 weeks. ALE was administered in dose of 1 mg/kg once a week. Every 4 weeks 8 rats from each group were sacrificed and their blood and both tibiae were harvested. The expression of osteocalcin(OC) and C-terminal cross linked telopeptides of type Ⅰ collagen(CTX) in serum was measured by enzyme-linked immunosorbent assay (ELISA) and the tibiaes were subjected to metaphyseal three-point bending and micro-CT analysis. Result: OC rose after ovariectomy and was not appreciably changed by either ALE or WBV alone or in combination. ALE treatment significantly prevented an increase of CTX. Compared with the OVX+WBV group, nearly all tested indices such as the BV/TV, TV apparent, Tb.N, Tb.Th, and Conn.D were higher in the OVX+ALE group at week 12. At week 12, tibiae treated with ALE exhibited a significantly higher Fmax, energy absorption and stiffness compared to the OVX+WBV group, but there was no significant difference. Conclusion: Compared with the WBV, ALE was more effective at preventing bone loss and improved the trabecular architecture.
