      摘要 目的:研究深部脑刺激(DBS)对脑外伤(TBI)昏迷大鼠前额叶皮层γ-氨基丁酸(GABA)b受体表达变化的影响及促醒可能机制。 方法:将54只成年标准大鼠随机分为空白组、假刺激组、刺激组,应用DBS治疗TBI昏迷大鼠,观察其行为学变化,实验完成后12h处死每组大鼠,用蛋白质印迹法(WB)及免疫荧光技术检测前额叶皮质区GABA b受体的表达。 结果:①刺激组18只大鼠中16只大鼠出现翻正反射,假刺激组18只中9只出现翻正反射;②WB检测显示,刺激组大鼠前额叶皮质区GABA b受体表达水平显著低于假刺激组(P<0.05);③免疫荧光检测显示,刺激组大鼠前额叶皮质区GABA b受体表达水平显著低于假刺激组(P<0.05)。 结论:DBS可提高TBI昏迷大鼠意识状态,其机制可能与下调前额叶皮质区GABA b受体表达有关。

Abstract:
      Abstract Objective: To investigate the changes of γ- aminobutyric acid (GABA) b receptor expression in prefrontal cortex of coma rats models after traumatic brain injury(TBI) with deep brain stimulation (DBS),and to explore the effects and possible mechanisms of DBS in coma and wake. Method: Standard rats were divided into 3 groups: control group, sham-stimulation group, stimulation group. DBS was applied to TBI coma rats. Then, the behaviors were observed and the prefrontal cortices of each group were extracted at 12h after the experiment. Western Blot(WB) and Immunofluorescence technique were used to detect the expressions of GABA b receptor. Result: Sixteen of 18 rats exhibited righting reflex in the stimulation group,and 9 of 18 rats had the same responses in the sham-stimulation groups.GABA b receptor levels in the stimulated groups were lower than the sham-stimulation groups in prefrontal cortex under WB(P<0.05). GABA b receptor levels in the stimulated groups were lower than the sham-stimulation groups in prefrontal cortex under Immunofluorescence technique(P<0.05). Conclusion:DBS could raise consciousness in TBI induced coma rats,and the potential mechanism may be related to the decreas of the expression of GABA b receptor in prefrontal cortex.
