      摘要 目的:观察超短波对大鼠急性肺损伤炎症反应的作用及核因子-κB(nuclear factor-κB,NF-κB)、Toll样受体4(Toll-like receptors 4, TLR4)表达的影响。 方法:将24只雄性SD大鼠随机分成3组:对照组,急性肺损伤组,超短波组,每组8只。气管内滴注脂多糖建立急性肺损伤模型。造模后的即刻、4h、8h分别给予干预组大鼠超短波治疗,电极板胸背对置,间距11cm,每次干预15min。采用HE染色观察肺组织病理学表现,肺损伤评分评估损伤程度,湿/干重比值(W/D)评估肺水肿,ELISA检测血清炎症因子白介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumor necrosis factor alpha, TNF-α)的水平,RT-PCR及Western-blot分别检测NF-κB、TLR4 mRNA及蛋白表达水平。 结果:与对照组比较,HE染色见急性肺损伤组大鼠肺组织结构受损,肺损伤评分及肺组织湿/干重比值(W/D)增高(P<0.01);血清炎症因子IL-6、TNF-α的水平上升(P<0.01);NF-κB、TLR4 mRNA表达及蛋白相对表达量明显升高(P<0.01)。与急性肺损伤组比较,超短波组大鼠肺组织结构受损程度明显减轻,肺损伤评分降低(P<0.01);肺组织湿/干重比值(W/D)下降,但尚不具备显著性意义(P>0.05);血清炎症因子IL-6、TNF-α水平下降(P<0.05,P<0.01);NF-κB、TLR4 mRNA表达及蛋白相对表达量降低(P<0.01,P<0.01)。 结论:超短波通过抑制NF-κB/TLR4信号通路,降低炎症因子IL-6、TNF-α的水平,抑制急性肺损伤炎症反应。

Abstract:
      Abstract Objective: To observe the effects of ultrashort wave on the inflammation of rats with acute lung injury and the expression of NF-κB and TLR4. Method: Twenty-four male SD rats were randomly divided into three groups: the control group, the acute lung injury group, and the ultrashort wave group, eight rats for each. The acute lung injury model was established by intratracheal injection of lipopolysaccharide. The rats in the ultrashort wave group were given ultrashort wave treatment immediately, 4h and 8h after lipopolysaccharide infusion. Battery lead plates were put symmetry between chest and back, keep eleven centimeters apart, each time for 15 minutes. The pathological changes in lung tissue of rats with acute lung injury were observed by HE staining, and evaluated by lung injury score. The wet/dry ratio(W/D) of lung tissue was used to evaluate the pulmonary edema. Serum levels of inflammatory factors IL-6 and TNF-α were detected by ELISA, mRNA and protein expressions of NF-κB and TLR4 were detected by RT-PCR and Western-Blot, respectively. Result: Compared with the control group, HE staining showed that the lung structure of rats in the acute lung injury group was damaged, lung injury score was significantly increased(P<0.01). Lung tissue wet/dry ratio(W/D)was increased(P<0.01). Serum levels of inflammatory factors IL-6 and TNF-α were increased(P<0.01). mRNA and protein expressions of NF-κB and TLR4 were significantly increased(P<0.01, P<0.01).Compared with the acute lung injury group, the damage degree of lung tissue structure in the rats of the ultrashort wave group was significantly reduced, lung injury score reduced(P<0.01), lung tissue wet/dry ratio(W/D) was also reduced, but the difference was not statistically significant(P>0.05).Serum levels of inflammatory factors IL-6 and TNF-α were decreased(P<0.05,P<0.01). mRNA and protein expressions of NF-κB and TLR4 were significantly decreased(P<0.01, P<0.01). Conclusion: Ultrashort wave regulates the NF-κB/TLR4 signal pathway to reduce the level of inflammatory factors IL-6 and TNF-α, and suppresses the inflammation response in the rats with acute lung injury.
